Hello, new here, I am going to put up some large lots of plush toys, and would like to know if any one knows the best way to ship them.

That is a pretty vague question!  Are they children's toys - unjointed play bears?  Are you only shipping in the US?  If so the answer really lies in what your buyers want - parcel post, UPS ground, and Fed Ex are the best ways to ship larger packages.  Pack them in plain brown boxes and weigh them, and then enter the weight when you list and offer at least UPS and parcel post USPS as options.  I would pack them in inexpensive trash bags to protect them from any dampness in case the shipping box gets wet.  I also put something over the bears - a piece of foam or bubble wrap - on top where a buyer might normally use a knife to cut the packing tape - so they don't cut into the bears - and put a warning on top to be careful cutting.
